Antiseizure efficacy of sodium channel blocking agent carbamazepine (CBZ) in a rodent model of amygdala kindling is not known. Methods ======= In anaesthetised rats, parietal cortical field potentials (FPs) were elicited by stimulating the perforant pathway and the evoked FP responses were recorded in the amygdala. After 5 min of baseline recording, amygdala kindling was induced by stimulating the amygdala. The FP recording was continued for 15 min after discontinuation of the amygdala stimulus trains. The CBZ-induced changes in FP were recorded and expressed as the percentage change in average FP amplitude relative to baseline. The dose of CBZ was 1.0 mg/kg. Statistical significance was calculated using ANOVA and Bonferroni's test for *post hoc*comparison. Results ======= The kindled FP responses increased progressively during the period of the amygdala stimulus train. The test drug, CBZ, when administered 10 min after the amygdala stimulus trains, was unable to change the kindled FP in the amygdala. On the other hand, CBZ, when administered 15 min after the amygdala stimulus trains, attenuated the FP responses in the amygdala. This effect of CBZ was dose-related, as the lowest dose, 0.5 mg/kg, did not induce any effect while higher dose of 1.0 mg/kg was able to attenuate the FP amplitude. Conclusion ========== The data suggest that CBZ, administered at lower dose, may be able to attenuate the development of amygdala kindled seizure and the sodium channel effects of anaesthetic drugs may be blocked at higher doses.
